摘要: Profilin has recently been identified as an actin-binding protein in higher plants. A cDNA coding for tobacco profilin, which shared average sequence identity of 75% with other plant profilins, was isolated from a pollen library by antibody screening. Tobacco profilin expressed Escherichia coli and purified affinity to poly-(L-proline) Sepharose. rabbit antiserum raised against recombinant used estimate the amount different tissues. can be detected somatic tissues, but expression is 50–100 fold mature pollen. Immunofluorescence confocal laser scanning microscopy showed homogeneous distribution cytoplasm vitro cultured grains tubes whereas some growing were stained more intensively their tip. possible role developmentally upregulated microfilament precursor discussed.
