摘要: Bone erosion is a sign of severe rheumatoid arthritis and osteoclasts play major role in the bone resorption. Recently, myeloid-derived suppressor cells (MDSC) has been reported to be increased collagen-induced (CIA). The number circulating MDSCs shown correlate with arthritis. These findings suggest that are precursor involved erosion. In this study, isolated from mice CIA stimulated M-CSF RANKL vitro expressed osteoclast markers acquired resorption function. sorted were transferred into tibia normal DBA/1J bones subjected histological Micro CT analyses. CIA-MDSCs differentiate TRAP(+) capable vivo. had more potent activity much less capability osteoclast. Additional experiments showed NF-κB inhibitor Bay 11-7082 or IκB peptide blocked differentiation IL-1Ra also differentiation. contrast, addition IL-1α further enhanced results source precursors inflammatory cytokines such as IL-1, contributing significantly erosive changes seen related disorders.
