摘要: Arachidonylethanolamide (anandamide) was identified some 15 years ago as a brain constituent that binds to the cannabinoid receptor. After this seminal discovery, multiple new receptors for anandamide have been identified, including vanilloid receptor (TRPV1), and is now frequently referred an "endovanilloid." Characterization of action on TRPV1 revealed (1) potency efficacy very much depend species tissue, (2) responsiveness in vivo significantly controlled by its local metabolism, (3) activation regulates responsiveness, (4) synthesis, (5) metabolites affect responses, (6) often observed convergent physiological actions agonists neither case necessarily represent direct effects TRPV1, (7) coactivation complicates distinction between these pathways. These issues are reviewed here together with potential implications pathophysiological pharmacological regulation inflammatory, respiratory, cardiovascular disorders, well appetite fat metabolism.
