Methylglyoxal-derivative advanced glycation endproducts: detection by competitive immunofluorometric assay and quantifying in serum and urine.

摘要: BACKGROUND Advanced glycation endproducts (AGE) are a family of heterogeneous chemical structures formed on the host protein in conditions carbonyl or oxidative stress. Among AGE precursors, methylglyoxal (MG) is considered one key intermediates. METHODS In current study, we describe and evaluate solid phase time-resolved fluoroimmunoassay (DELFIA) based competitive reaction between MG-AGE antibody antigen for detecting MG-adducts serum urine. The fluorometry assay was validated by comparison with previously established ELISA. RESULTS concentration assayed DELFIA sera diabetic patients (DM, n=66) were higher to non-diabetic controls (C, n=28); DM median=294 mgEq/L (10th 90th percentile 158-564) vs. C median=224 124-290) (p=0.0022). subjects, urinary excretion exceeded mean level measured controls; median=36.0 1-210) median=11.5 1-49) p=0.0036. urine low undetectable 8 28 control subjects 2 66 diabetics. percentage recovery MG-adduct added same pool showed under-recovery latter. Comparison total amount revealed MG-derivatives account 37% structure commonly termed AGE. CONCLUSIONS MG-derivative evaluated can be utilized biomonitoring its excretion. found substantially more sensitive than standard ELISA having wider dynamic range, while sharing similar quenching attributes.
