Oleic acid Enhances Dengue Virus But Not Dengue Virus-Like Particle Production from Mammalian Cells

摘要: Despite the recent introduction of a commercial vaccine, mosquito-transmitted dengue virus is still worldwide public health problem. Based on live attenuated vaccine strategy, has less than optimal protective profile. Virus-like particles (VLPs) offer an attractive alternate vaccination strategy due to effectively native presentation epitopes in absence any infectious genetic material. However, production amounts VLP platform that can support development remains major obstacle. This study generated two DENV 2 VLPs [codon-optimized and chimeric DENV/Japanese encephalitis (JEV)] directly compared yields these constructs by western blotting dot blot hybridization. The effect oleic acid supplementation, process known increase natural infection, was also investigated. Results showed construct gave two- threefold higher yield codon-optimized while increased virion it inhibited production. These results suggest further optimization expression possible, but will require more understanding how infection remodels host cell machinery.
