摘要: We have developed a highly sensitive and selective method for the determination of Leukotriene B4 (LTB4) in human plasma using negative ion chemical ionization/gas chromatography/tandem mass spectrometry (NICI/GS/MS/MS) analysis. The was summarized as follows. Deuterated LTB4 (d4-LTB4) added to samples an internal standard, were extracted by Sep-pak C18 column. Extracted derivatized into pentafluorobenzyl ester bis-trimethylsilyl ether (PFB-TMS-LTB4) quantified on basis selected reaction monitoring (SRM) at m/z 299 [M-PFB-2TMSOH]- NICI/GC/MS/MS analysis, which product [M-PFB]-. detection limit quantification 10 pg ml-1, sufficiently determine concentrations endogenous plasma. level measured healthy male volunteers 33.85 +/- 33.91 ml-1 (mean S.D. six volunteers). technique MS/MS used this offers much greater sensitivity selectivity than single-stage spectrometry. showed good reproducibility with simple rapid extraction procedure, would be useful examining relationship between various disease states levels biological fluids.
