Preliminary evaluation of technetium-99m-labeled ceftriaxone: infection imaging agent for the clinical diagnosis of orthopedic infection.

摘要: Summary Objective In this study we sought to assess the efficacy of a technetium-99m (Tc-99m)-labeled third-generation cephalosporin as an infection imaging agent in accurate detection sites bacterial vivo. Design Ceftriaxone (CRO) was formulated into ready-to-use single-vial cold kit with shelf-life over 6 months and successfully labeled technetium. The radiolabeled drug, Tc-99m-CRO, subjected following preclinical evaluations: radiochemical purity, vitro vivo stability, binding assay, pharmacokinetic studies animals human patients. Results formulation exhibited excellent radiolabeling efficiency (∼99%) high stability. drug slow blood clearance (12% at 4 h), protein excretion pattern mimics reported profile alone. animal model, scintigraphy scans showed higher uptake radiopharmaceutical infectious lesions, even 1 h post-administration, comparison inflammatory lesions. clinical evaluation Tc-99m-labeled CRO diagnostic accuracy 83.3%, sensitivity specificity 85.2% 77.8%, respectively. Conclusions This has potential for infections much compared other antibiotics available convenient kits routine practice.
