Cyclic AMP treatment of Rous sarcoma virus-transformed Chinese hamster ovary cells increases phosphorylation of pp60src and increases pp60src kinase activity.

摘要: Abstract Treatment of growing Rous sarcoma virus-transformed Chinese hamster ovary cells with the cyclic AMP analog 8-bromo-cyclic adenosine 3',5'-monophosphate (8-bromo-cyclic AMP) stimulates incorporation 32Pi into viral transforming protein pp60src. Based on one-dimensional and two-dimensional peptide analysis phosphoamino acid analysis, increase is a single phosphoserine residue at NH2 terminus protein. The phosphate increases during first 4 h treatment. pp60src kinase activity in extracts treated was stimulated about 2- to 3-fold. This stimulation increased 3 treatment 1 mM 8-bromo-cAMP both soluble particulate fraction cells. These results suggest that can modulate transformed
