摘要: Maturation of protein N-termini occurs in all kingdoms life, with major modifications being proteolytic processing (e.g., removal initiator methionines) and N-terminal acetylation. The functional consequences these are only known for a few substrates, techniques to study such have begun emerge recently. We here report on method enabling targeted, mass spectrometry based analysis from polyacrylamide gel-separated proteins. In our method, stable isotope incorporation by in-gel N-acetylation free primary amines permits calculating the extent vivo acetylation, proven reveal crucial information reference biology.
