摘要: Objective: To investigate whether the umbilical cord mesenchymal stem cells (UC-MSCs) transplantation in MRL/lpr mice has effect or not on their pneumonitis and possible mechanisms underlying this treatment. Methods: Twenty four 18-week-old female were divided into three groups as following: group 2 (UC-MSCT group) have been transplanted with 1×106 UC-MSCs through caudal vein, 3 (multi-UC-MSCT Group) times 1 (control treated 0.5 mL phosphate buffer saline (PBS) control. The histopathology of lung was observed. pulmonary expression high mobility box protein-1 (HMGB-1) measured by western blot detected quantitation real time polymerase chain reaction (PCR). Immunohistochemistry method used to detect HMGB-1 expressions pulmo. Results: In comparision control ground mice, significantly reduced interstitial mice. peribronchiolar lesion index UC-MSCT (1.40±0.24) multi-UC-MSCT (1.02±0.29) decreased compared (1.95±0.35) (P<0.01). perivascular (1.20±0.18) (1.08±0.16) also (1.56±0.32) (P=0.018, 0.002) alveolar areas (1.72±0.34) increased (1.30±0.21) (1.05±0.15) (P=0.011, 0.000). protein (0.32±0.04) (0.28±0.06) both that (0.80±0.21) (P<0.05). level mRNA (4.68±0.37) (4.35±0.10) those (16.29±3.84) immunohistochemical staining sections, found mainly located cytoplasm extracellular matrix MRL/ lpr epithelial cells, group. Conclusions: These findings indicate a therapeutic systemic lupus erythematosus (SLE) pneumonitis, possibly inhibiting expression, which suggests potential application treatment human lupus.
