J Periodont Res 2012; 47: 645–654. © 2012 John Wiley & Sons A/S Background Objective: Recombinant (rhBMP-2) is a well-known growth factor that can induce robust formation, recent studies have shown rhBMP-2-induced osteogenesis closely related to adipogenesis. The aim present study was determine dose- rhBMP-2 differentiation cells (hABCs) in vivo vitro. Material Methods: hABCs were isolated cultured, then transplanted using carrier treated either with or without (100 μg/mL) into an ectopic subcutaneous mouse model. Comprehensive histologic histometric analyses performed after 8-wk healing period. To further understand dose-dependent (0, 10, 50, 200, 500 1000 ng/mL) 3, 5, 7 14 d) differentiation, vitro evaluated, expression mRNAs, including those for alkaline phosphatase, osteocalcin, sialoprotein, peroxisome-proliferator-activated receptor gamma-2 lipoprotein lipase, assessed quantitative RT-PCR. Results: significantly promoted vivo, gradually increased both potential manner minimal deviation vitro. osteogenesis- adipogenesis-associated mRNAs concomitantly up-regulated by rhBMP-2. Conclusion: findings showed enhanced as well manner. control should be considered when regenerating rhBMP-2.
