Germ-line transformation of pink bollworm (Lepidoptera: Gelechiidae) mediated by the piggyBac transposable element.

摘要: The pink bollworm, Pectinophora gossypiella, is a world-wide pest of cultivated cotton. In certain growing regions populations are suppressed by sterile release strategy. Efforts to improve the insect technique as well our understanding lepidopteran biology could benefit greatly from germ-line transformation system. We report bollworm with piggyBac transposable element carrying enhanced green flourescent protein (EGFP) marker gene. This vector-marker system resulted in recovery transgenics at rate approximately 3.5%. Integration transforming construct that was typical demonstrated Southern analysis and sequence determination transposon flanks. Expression EGFP visualized fluorescent microscopy Western Blot analysis. Maintenance transformed strains indicates transgene segregates Mendelian fashion has been stable over fourteen generations date.
