Modulation of Dcytb (Cybrd 1) expression and function by iron, dehydroascorbate and Hif-2α in cultured cells

摘要: Abstract Background Duodenal cytochrome b (Dcytb) is a mammalian plasma ferric reductase enzyme that catalyses the reduction of to ferrous ion in process iron absorption. The current study investigates relationship between Dcytb, iron, dehydroascorbate (DHA) and Hif-2α cultured cell lines. Methods Dcytb protein expression was analysed by Western blot technique while gene regulation determined quantitative PCR. Functional analyses were carried out 59Fe uptake assays. Results Iron dehydroascorbic acid treatment cells inhibited mRNA expression. Desferrioxamine also enhanced level after treated overnight. knockdown HuTu resulted reduced lowered activity. Preloading with DHA (to enhance intracellular ascorbate levels) did not stimulate activity fully Dcytb-silenced cells, implying Dcytb-dependence ascorbate-mediated ferrireduction. Moreover, led uptake. Conclusions Taken together, this shows functional Hif-2α. General significance membrane accepts electrons intracellularly from DHA/ascorbic for ferrireduction at apical surface enterocytes.
