摘要: PURPOSE. To investigate the role of protein kinase C (PKC) in cholinergic agonist-induced Ca 2+ elevation lacrimal gland acini. METHODS. Lacrimal acini were prepared by collagenase digestion, and changes intracellular 2 ([Ca ] i ) measured using fura-2 as a fluorescent probe. RESULTS. Preactivation PKC phorbol 12-myristate 13-acetate (PMA), or inhibition phosphatase type 1/2A (PP1/2A) calyculin A, decreased both [Ca transient plateau induced increasing concentrations carbachol, agonist. Staurosporine, an inhibitor PKC, completely reversed effect PMA. Inhibition -independent isoforms PKCδ -e, but not -dependent isoform PKCα substantially inhibitory PMA on elevation. The was obtained only presence extracellular , suggesting that inhibits influx . inhibited A thapsigargin, further supporting idea modulates entry CONCLUSIONS. In gland, are negatively regulated PKC-dependent phosphorylation target protein(s) is sensitive to PP1/2A.
