Regulation of skeletal growth and mineral acquisition by the GH/IGF-1 axis: Lessons from mouse models

摘要: The growth hormone (GH) and its downstream mediator, the insulin-like factor-1 (IGF-1), construct a pleotropic axis affecting growth, metabolism, organ function. Serum levels of GH/IGF-1 rise during pubertal associate with peak bone acquisition, while aging their decline loss. was extensively studied in numerous biological systems including rodent models cell cultures. Both hormones act an endocrine autocrine/paracrine fashion understanding distinct overlapping contributions to skeletal acquisition is still matter debate. GH IGF-1 exert effects on osteogenic cells via binding cognate receptor, leading activation array genes that mediate cellular differentiation interact other regulators, such as sex-steroids, thyroid hormone, parathyroid facilitate metabolism. In this review we summarized several described key experiments shed new light regulation by axis.
