Gene Targeting in Mice: a Review

摘要: The ability to introduce DNA sequences (e.g., genes) of interest into the germline genome has rendered mouse a powerful and indispensable experimental model in fundamental medical research. can be integrated randomly or specific locus by homologous recombination, order to: (1) delete insert mutations genes determine their function, (2) human mice generate animal models enabling study human-specific diseases, e.g., susceptible infections pathogens interest, (3) individual genomes (such as viruses) examine contributions such pathogenesis parent pathogens, (4) last but not least reporter that allow monitoring vivo ex expression interest. Furthermore, use recombination systems, Cre/loxP FRT/FLP, enables conditional induction suppression gene restricted period mouse's lifetime, particular cell type, tissue. In this review, we will give an updated summary targeting technology discuss some important considerations design gene-targeted mice.
