摘要: Abstract Ethnopharmacological relevance Pogostemon cablin (Blanco) Benth is a well-known medicinal herb commonly used in many Asian countries for inflammatory diseases. Pogostone (PO), natural product isolated from , known to exert various pharmacological activities. This study aimed investigate the anti-inflammatory property of PO, elucidate its mechanism action, and evaluate potential acute toxicity. Materials methods The vitro activity PO was assessed using lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. protein mRNA levels proinflammatory mediators were measured with ELISA RT-PCR, respectively. Proteins NF- κ B MAPK family determined by Western blot underlying molecular mechanisms. vivo tested LPS-induced endotoxic shock mice. In addition, median lethal dose (LD 50 ) mice an toxicity test. Results vitro, significantly inhibited expression including TNF-α, IL-6, IL-1β, NO, PGE 2 . action partly dependent on inhibition activation phosphorylation JNK p38 MAPK. able reduce mortality induced LPS Furthermore, could markedly suppress production serum, attenuate liver lung injury. mechanisms during may be attributed down-regulation multiple organs via Moreover, LD about 163 mg/kg intravenous administration, which 8-fold higher than animal experiment. Conclusions Our findings regarding effect help justify use Chinese medicine treatment More importantly, results also render promising agent worthy further development into pharmaceutical drug septic shock.
