摘要: In fission yeast, overexpression of the replication initiator protein Cdc18p induces re-replication, a phenotype characterized by continuous DNA synthesis in absence cell division. contrast, Cdc6p, budding yeast homolog Cdc18p, does not cause re-replication S. cerevisiae. However, we have found that Cdc6p has ability to induce yeast. cannot functionally replace but instead interferes with proteolysis both and Rum1p, inhibitor kinase Cdc2p. This activity is entirely contained within short N-terminal peptide, which forms tight complex Cdc2p F-box/WD-repeat Sud1p/Pop2p, component SCFPop ubiquitin ligase These interactions are mediated two distinct regions region depend on integrity its phosphorylation sites. The data suggest disruption control consequence sequestration Pop2p, factors involved negative regulation potentially other proteins.
