摘要: The organization of immunoglobulin heavy (H) chain genes in teleosts resembles that mammals and amphibians, whereas light (L) are arranged multiple clusters variable (VL), joining (JL), constant (CL) region segments. Sequence analysis two Atlantic cod genomic clones (14,966 13,116 bp length) revealed a very compact IgL locus with the VL opposite transcriptional orientation to JL CL genes. This suggests possibility rearrangements between by inversion. Each cluster spans approximately 2.1 kb distances vary 4.8 kb. To gain insight into regulation this complex, multiclustered locus, chloramphenicol acetyl transferase reporter constructs containing 14 different DNA segments from were transfected channel catfish B non-B-cell lines, as well mouse B-cell lines. These studies showed strong enhancer activity downstream three out six L gene when assayed fish, but not cells. Interestingly, both human λ enhancers exhibited fish cells, while 3′κ did not. transcription factors similar those involved mammalian expression present cells teleosts.
