Characterization of the cellulolytic complex (cellulosome) of Clostridium acetobutylicum

摘要: A large cellulosomal gene cluster was identified in the recently sequenced genome of Clostridium acetobutylicum ATCC 824. Sequence analysis revealed that this contains genes for scaffolding protein CipA, processive endocellulase Cel48A, several endoglucanases families 5 and 9, mannanase Man5G, a hydrophobic protein, OrfXp. Surprisingly, genetic organization is very similar to cellulolyticum, model mesophilic clostridial cellulosomes. As C. unable grow on cellulosic substrates, existence raises questions about its expression, function evolution. Biochemical evidence expression complex investigated. The results sodium dodecyl sulfate–polyacrylamide gel electrophoresis, N-terminal sequencing Western blotting with antibodies against specific components cellulolyticum cellulosome suggest at least four major proteins are present. In addition, despite fact no cellulolytic activities were detected, we report here production high molecular mass acetobutylicum.
