摘要: Background: The utilization of stem cell trans-differentiation into insulin-producing cells (IPCs) would provide potential promising therapy for diabetes mellitus (DM). Objective: study was aimed to investigate the differentiation rabbit’s bone marrowderived insulin producing cells. Methods: Bone marrow-derived mesenchymal (MSCs) were obtained from three New-Zealand male white rabbits and propagated in a primary culture 14 days low glucose Dulbecco's Modified Eagle's Medium (DMEM), harvested subjected another passages encompassing 21 days. After that, MSCs functionally defined by their ability differentiate osteoblasts. This achieved incubation with DMEM containing 10 -7 M dexamethasone, mM βglycerophosphate 50 μl/ml ascorbic acid. Osteogenic followed up at 2, 4, 7 9 staining alizarin red S vonKossa. Results: It found that onset day continued 9. On other hand, patch induced two step incubation. first high serum-free 0.5 mmol/L β-mercaptoethanol second follows same medium nicotinamideinstead β-mercaptoethanolfor 18 Trans-differentiation 9, 21. have trans-differentiated starting subsequent as judged affinity stain diphenylthiocarbazone (dithizone or DTZ) Conclusion: results some insights using rabbit's marrow source potentials. might help development future well several human diseases.
