摘要: Resistance to many intestinal nematodes is dependent on the induction of polarized type 2 cytokine responses, whereas 1 responses can exacerbate these infections. The contributions IL-4 and IL-13 development resistance have been well described for a variety parasites; however, role IL-10 has not previously investigated. In this study we infected IL-10-, IL-10/IL-4-, IL-10/IL-12-, IL-4-, IL-12-deficient mice with Trichuris muris determine whether contributes immunity. Interestingly, T. muris-infected IL-10/IL-4-deficient failed expel parasite, animals deficient in displayed marked morbidity mortality. contrast, double IL-10/IL-12-deficient were completely resistant mounted highly response, demonstrating that increased susceptibility IL-10-deficient was IL-12. Further suggested IL-10- probably attributable increase production those animals. mortality observed correlated inflammation, loss Paneth cells, absence mucus cecum. survival enhanced if broad spectrum antibiotic administered, suggesting an outgrowth opportunistic bacteria contributing high degree Overall, studies reveal critical polarization Th2 during infection, maintenance barrier function colon.
