摘要: It has been shown that the association of opioids analgesic agents with non-steroidal anti-inflammatory drugs (NSAIDs) can increase their antinociceptive activity, allowing use lower doses and thus limiting side effects. Therefore, goal present study was to examine possible pharmacological interaction between acemetacin two in Hargreaves model thermal hyperalgesia mouse. Acemetacin, codeine, nalbuphine or fixed-dose ratios acemetacin-codeine acemetacin-nalbuphine combinations were administrated systemically mice antihyperalgesic effect evaluated using test. All treatments produced a dose-dependent effect. ED40 values estimated for all an isobologram constructed. The derived theoretical 55.9+/-4.9 mg/kg 40.3+/-3.8 mg/kg, respectively, being significantly higher than actually observed experimental ED40, 14.5+/-1.7 12.7+/-2.2 respectively. These results correspond synergistic interactions on hyperalgesia. Highest individual did not affect motor coordination balancing test rota-rod. Data suggest low acemetacin-opioids combination interact synergistically at systemic level therefore this may represent therapeutic advantage clinical treatment inflammatory pain.
