摘要: Differential targeting of neuronal proteins to axons and dendrites is essential for directional information flow within the brain, however, little known about this protein-sorting process. Here, we investigate polarized lipid-anchored peripheral membrane proteins, postsynaptic density-95 (PSD-95) growth-associated protein-43 (GAP-43). Whereas N-terminal palmitoylated motif PSD-95 necessary but not sufficient sorting dendrites, palmitoylation GAP-43 axonal can redirect a chimera axons. Systematic mutagenesis motifs indicates that spacing cysteines presence nearby basic amino acids determine by these two motifs. Similarly, protein paralemmin contains C-terminal domain, which resembles also mediates targeting. These axonally targeted mediate detergent-insoluble glycolipid-enriched complexes in heterologous cells, suggesting possible role specialized lipid domains proteins.
