Critical role of TrkB and brain-derived neurotrophic factor in the differentiation and survival of retinal pigment epithelium

摘要: In the vertebrate eye, retinal pigment epithelium (RPE) and neural retina arise from a single layer of neuroectoderm. Factors influencing differentiation neurons have been identified; however, little is known about molecules directing RPE. Here we found that neurotrophin brain-derived neurotrophic factor (BDNF) plays an autocrine role in survival Xenopus laevis Fluorescent situ hybridization studies showed precise co-expression BDNF its receptor trkB neuroepithelium actively differentiating RPE; vitro demonstrated survival- differentiation-promoting effects serum-free explants dissociated cultures. When dominant negative mutant was expressed developing embryos, severe arrest RPE seen with persistence nestin- Notch-positive neuroblasts.
