Morphoproteomic confirmation of an activated nuclear factor-кBp65 pathway in follicular thyroid carcinoma.

摘要: The role of nuclear factor (NF)-кBp65 pathway in the pathogenesis follicular thyroid carcinoma (FTC) has not been fully investigated. We retrieved 10 cases FTC from our file. Tissue microarrays (TMAs) were constructed using 2.0 mm cores formalin-fixed, paraffin-embedded tissue blocks. TMA sections immunohistochemically stained for phosphorylated (p)-NF-кBp65 (Ser 536), cyclooxygenase-2 (COX-2), IL-8, and glutathione S-transferase (GST)-pi. Staining intensity (0-3+), extensiveness (0-100%) subcellular compartmentalization evaluated. Both cytoplasmic immunoreactivities with p-NF-кBp65 536) antibodies observed all cases, including moderate to strong staining a range 20% - 100% tumor cells. Moderate (2+) or (3+) expressions COX-2 IL-8 present 60-100% 50- cells, respectively, cases. GST-pi was diffusely (70-100%) moderately strongly cytoplasm (except one case insufficient tissue) three them demonstrating positivity as well. Morphoproteomic analysis reveals constitutive activation NF-кBp65 carcinomas evidenced by phosphorylation at Ser 536 translocation correlative expression transcriptionally activated gene products (COX-2, GST-pi). This observation may provide molecular basis biology targeted therapies carcinoma.
