Synthèses enzymatiques de néoglucoconjugués catalysées par l'alpha-glucosidase purifiée de la blatte Periplaneta americana (Linnaeus)

摘要: La blatte Periplaneta americana (Linnaeus) possede dans son tube digestif une alpha-glucosidase mesophile (50 °C) ayant activite hydrolytique maximale a pH 5,0. Cette enzyme, purifiee homogeneite electrophoretique sur gel de polyacrylamide 12 % (P/V), hydrolyse activement le maltose, saccharose et p-nitrophenyl-alpha-D-glucopyranoside. Nous avons teste l’aptitude cette enzyme catalyser les reactions synthese neoglucoconjugues lorsque maltose sont utilises comme donneurs glucosyle, 2-phenylethanol phenol accepteurs glucosyle (aglycones). Les conditions experimentales ont ete optimisees au niveau du temps reaction, des concentrations d’accepteurs glucosyle. pourcentages reaction transglucosylation [donnes par rapport quantite synthetises (mM)/quantite produit d’hydrolyse (mM)] 37 °C tres eleves pu atteindre 67 % 48 % respectivement avec produits ceux-ci deviennent importantes milieu reactionnel. Ce qui signifie que ces phenylethyl-alpha-D-glucoside phenyl-alpha-D-glucoside. Ces resultats suggerent fonctionne un mecanisme retention configuration anomerique. A partir ce constat, l’alpha-glucosidase la apparait bonne source pour preparation alpha-neoglucoconjugues.
