Hypoxia Enhances the Expression of Follistatin-like 3 in Term Human Trophoblasts☆

摘要: Hypoxic injury hinders placental differentiation and alters trophoblast gene expression. We tested the hypothesis that expression of follistatin-like 3 (FSTL3), a member follistatin family proteins, is modulated by hypoxia in primary human (PHT). Using immunofluorescence term villi we detected FSTL3 protein villi, primarily trophoblasts. verified this finding cultured PHT cells. Basal transcript cells, determined using quantitative PCR, was stable over culture period. Importantly, when compared to FiO2=20% or FiO2=8%, cells FiO2 <1% exhibited 4–6 fold increase mRNA as early 4 h hypoxia. Whereas cellular unchanged hypoxia, found increased level medium. Lastly, exposure either hypoxia-mimetic cobalt chloride proline hydroxylase inhibitor dimethyloxaloylglycine upregulated transcript. Our data indicate enhances its release from agents enhance implicates HIF1α process.
