Optimized conditions for high erythritol production byPenicillium sp. KJ-UV29, mutant ofPenicillium sp. KJ81

摘要: To improve the erythritol productivity ofPenicillium sp. KJ81, mutants were obtained using UV irradiation and NTG treatment. Among these mutants,Penicillium KJ-UV29 revealed no morphological changes, yet was superior to wild strain in following three points: (1)Penicillium produced more than under same conditions, (2) foam during cultivation, unlike strain, (3) mutant a significantly lower amount of glycerol.Penicillium sp as much 15.1 g/L erythritol, whereas wild-typePenicillium KJ81 only 11.7 g/L.Penicillium generated 6.1 glycerol, compared 19.4 by strain. When investigating optimal culture conditions for production strainPenicillium KJ-UV29, sucrose idetified most effective carbon source, even able produce 70% sucrose-containing medium, although 30% medium exhibited highest productivity. The byPenicillium also increased addition ammonium carbonate, potassium nitrate, sodium nitrate. Accordingly, conditions,Penicillium 45.2 containing sucrose, 0.5% yeast extract, (NH4)2C2O4 0.1% NaNO3, 0.01% FeSO4 with 1 vvm aeration 200 rpm agitation at 37°C 7 days 5-L jar fermentor.
