Epidermal growth factor receptor and HER-2/neu status by immunohistochemistry and fluorescence in situ hybridization in adenocarcinomas of the biliary tree and gallbladder.

摘要: Adenocarcinomas of the biliary tract and gallbladder are aggressive tumors with a poor prognosis. Standard chemotherapy often offers minimal benefit. Because epidermal growth factor receptor HER-2/neu antagonists have been successfully used in adenocarcinomas from other sites, their use cholangiocarcinoma can be potentially beneficial. This study examines expression gene copy number adenocarcinomas. Fifty-one formalin-fixed, paraffin-embedded cases (26 intrahepatic, 19 extrahepatic, 6 gallbladder) were stained monoclonal antibodies against HER-2/neu. Fluorescence situ hybridization analysis was performed 37 using probes directed centromeric region chromosome 7. Epidermal present 41 (80%) cases, moderate or strong staining 30 (59%) cases. positive 2 (4%) showed gain 17 (46%) tumors. Of latter, 1 amplification, whereas all others 7, indicating balanced polysomy. overexpression by immunohistochemistry correlated significantly fluorescence (P = .02). HER2/neu is uncommon these
