摘要: In order to identify a promoter system for high-level expression of transgenes in hairy roots, we characterized the chimeric super-promoter fused translational enhancer from tobacco etch virus (TEV). Transgenic plants and roots were generated with super-promoter:TEV sequence modified green fluorescence protein (mGFP5) as reporter gene. To exploit utility root cultures secretion-based system, signal peptide patatin was mGFP5 direct its secretion into culture medium. Levels RNA on average sixfold higher than leaves. Likewise, GFP levels per gram fresh weight at least tenfold Furthermore, more 10% recombinant produced found Immunoblotting anti-GFP antibodies showed two products 27.1 29.9 kDa all leaf tissue extracts, whereas single 27.1-kDa product detected Inducibility studied mature leaves 14-day (midlog phase) roots. A twofold increase mRNA immediately after wounding both corresponding 24 h. Our studies demonstrate proteins bioreactors.
