摘要: The family of G-protein-coupled receptors can be defined by their similar structural and functional characteristics. Although primary sequences are quite diverse, these proteins share several common features that reflect mechanism action. Mutagenesis biophysical analysis indicate small molecule agonists antagonists bind to a hydrophobic pocket buried in the transmembrane core receptor. In contrast, peptide ligands both extracellular domains. mechanisms which cause receptor activation being explored various approaches, but not yet well defined. A deeper understanding basis for activity this large will have important implications drug design variety therapeutic areas.
