Poly I:C facilitates the phosphorylation of Ctenopharyngodon idellus type I IFN receptor subunits and JAK kinase.

摘要: Members of the Janus kinase (JAK) family, JAK1 and TYK2 take part in JAK-STAT signaling pathway mediated by interferon mammalian cells. Similar to counterparts, fish also perform their potential biological activities phosphorylating cytokine receptors STAT. In present study, Ctenopharyngodon idellus (CiJAK1) (CiTYK2) were cloned identified. The full-length cDNA CiJAK1 (KT724352.1) is 3829 bp, with an Open Reading Frame (ORF) 3465 bp encoding a putative protein 1154 amino acids. CiTYK2 (KT724353.1) 4337 including ORF 3168 1055 Structurally, both them have B41, SH2, TyrKc common domains. share high degree homology respective counterparts from Danio rerio Cyprinus carpio phylogenetic tree analysis. Polyinosinic-polycytidylic acid (Poly I:C), synthetic dsRNA analogue, can launch antiviral pathway. To elucidate molecular mechanism Poly I:C initiating fish, C. idellus kidney (CIK) cells stimulated then cell lysates separated on 10% SDS-PAGE. results showed that not only drastically increased expression level CiTYK2, but it induced phosphorylation as well type I IFN receptor subunits, CiCRFB1 CiCRFB5. detail, levels p-CiJAK1 p-CiTYK2 evidently up-regulated at 3 h post stimulation; however CiCRFB5 displayed sharp up-regulation 12 h stimulation I:C. As basic mechnism feedback regulation pathway, overexpression CIK facilitated CiTYK2.
