摘要: We describe an assay which uses broad-spectrum, conserved-site PCR paired with mass spectrometry analysis of amplicons (PCR/electrospray ionization-mass [ESI-MS]) to detect and identify diverse bacterial Candida species in uncultured specimens. The performance the was characterized using whole-blood samples spiked low titers 64 6 representing breadth coverage assay. had average limit detection 100 CFU bacteria or per milliliter blood, all tested yielded limits between 20 500 milliliter. Over 99% detections correct identifications, whether they were obtained at concentrations well above lowest detectable concentrations. This study demonstrates ability broad-spectrum PCR/ESI-MS assays organisms complex natural matrices that contain high levels background DNA.
