Acid-induced unfolding of myoglobin triggered by a laser pH jump method

摘要: Using 1-(2-nitrophenyl)ethyl sulfate (caged sulfate) as a photoactivatable caged proton, we could induce complete acid unfolding of myoglobin with single nanosecond laser pulse. This was possible because the high (≈ mM) concentration protons released by photolabile compound. The ability compound to produce large pH jump arises other photoproducts (2-nitrosoacetophenone and ion) do not buffer protons. time course kinetics, spanning range from milliseconds several seconds, be accurately reproduced monitoring absorbance changes in visible spectrum at 633 nm.
