摘要: Using retrovirus-mediated transfer of the SV40 virus large T antigen into neural transplants, we have observed a high incidence primitive neuroectodermal tumors (PNET). These neoplasms developed in 8 14 (57%) grafts after latency periods 176 to 311 days. Histopathologically, exhibited features human PNET such as formation neuroblastic rosettes and immunocytochemical evidence for neuronal differentiation, synaptogenesis, focal astrocytic differentiation. All showed striking migratory potential. The presence gene was demonstrated by polymerase chain reaction-mediated amplification specific 242 bp segment DNA sequence analysis. Large identified tissue sections using an reaction with monoclonal antibody Pab 108. Cell lines were established from several subjected G418 selection. Secondary induced intracerebral transplantation these cells retained characteristic morphological properties PNETs. experiments demonstrate considerable transforming potential precursor cells. long period suggests that neoplastic transformation initiated requires additional spontaneous mutations cooperating cellular genes. Because mechanism appears involve complex inactivation tumor suppressor products, cell may serve interesting tool search novel
