摘要: Hydroxyapatite (HA) coatings have been used to improve implant fixation by promoting bone formation around the prosthesis. A macrophage response HA particulates has noted loosened HA-coated prostheses. As biomaterial wear particle-associated macrophages are known be capable of differentiating into osteoclasts that resorption, we examined whether particulate could similarly induce macrophage-osteoclast differentiation. HA-associated were isolated from granulomas, formed subcutaneous implantation HA, and co-cultured with UMR 106 osteoblast-like cells in presence 1,25-dihydroxyvitamin D(3) for up 14 days on glass coverslips slices. differentiation was evidenced numerous multinucleated tartrate resistant acid phosphatase (TRAP)-positive which lacunar resorption pits Polymethylmethacrylate (PMMA) macrophages, PMMA-containing caused significantly more osteoclast than macrophages. These results indicate responding particles They also suggest derived uncemented (HA-coated) implants likely less osteolysis cemented implants.
