摘要: Tn antigen (Tn), a single N-acetylgalactosamine (GalNAc) monosaccharide attached to protein Ser/Thr residues, is found on most solid tumors yet rarely detected in adult tissues, featuring it one of the distinctive signatures cancers. Although prevalent cancers, Tn-glycosylation sites are not entirely clear owing lack suitable technology. Knowing will spur development new vaccines, diagnostics, and therapeutics Here, we report novel technology named EXoO-Tn for large-scale mapping sites. utilizes glycosyltransferase C1GalT1 isotopically-labeled UDP-Gal(13C6) tag convert Gal(13C6)-Tn, which has unique mass being distinguishable other glycans. This exquisite Gal(13C6)-Tn structure recognized by OpeRATOR that specifically cleaves N-termini Gal(13C6)-Tn-occupied residues yield site-containing glycopeptides. The use mapped 947 from 480 glycoproteins Jurkat cells. Given importance diseases, anticipated have broad utility clinical studies.
