Experimentally reduced insulin/IGF-1 signalling in adulthood extends lifespan of parents and improves Darwinian fitness of their offspring

摘要: Classical theory maintains that ageing evolves via energy trade-offs between reproduction and survival leading to accumulation of unrepaired cellular damage with age. In contrast, the emerging new postulates because deleterious late-life hyper-function reproduction-promoting genes excessive biosynthesis in late-life. The uniquely predicts optimizing nutrient-sensing molecular signalling adulthood can simultaneously postpone increase Darwinian fitness. Here we show reducing evolutionarily conserved insulin/IGF-1 daf-2 RNA interference (RNAi) fulfils this prediction Caenorhabditis elegans nematodes. Long-lived RNAi parents showed normal fecundity as self-fertilizing hermaphrodites improved when mated males. Remarkably, offspring had higher fitness across three different genotypes. Thus, reduced improves both parental longevity quality supporting view sub-optimal gene expression lies at heart ageing.
