The influence of genotype and medium on rye (Secale cereale L.) anther culture

摘要: Anthers of three rye inbred lines - L9, L318, Dw28, one synthetic hybrid F1(5) and variety, Dankowskie Zote (DZ), were cultured on two media based N6, P2 components. The induction rate significantly depended genotype the best results obtained for line L318 lowest percentage responding anthers was noticed in L9. There no universal medium all tested genotypes. highest (IR) L9 CI (11.94, 0.71 1.75 respectively). For DZ, P2I better than others while Dw28 CIP turned to be as suitable CI. A highly significant interaction between proved. Single produced embryos CI, media. They did not develop into plants but after transferring them CS1.7 medium, a secondary, embryogenic callus obtained. Such reaction has been described until now. In spite relatively high IR, plant regeneration rather poor. An elaboration this step haploid production is needed. Most analyzed calluses microspore derived regenerants proved haploids, according flow cytometry analysis. Plants treated with colchicine doubled haploids.
