摘要: Systemically administered lipopolysaccharide (LPS) elicits profound changes in pituitary hormone secretion. Pro-inflammatory cytokines have been proposed as mediators of these responses. In this study, we used in-situ hybridization histochemistry to investigate LPS-induced cytokine gene expression the rat pituitary. After i.p. or i.v. injection various doses LPS, mRNA for immediate-early IkappaBu (an inhibitor NF-kappaB, a transcription factor that regulates many pro-inflammatory cytokines) was induced anterior lobe early 0.5 h. The IkappaBalpha peaked at 1 posterior lobe, first h and 2 A similar spatiotemporal pattern interleukin-1b (IL-1) induction observed. addition, after injection, TNFalpha, IL-1beta converting enzyme (ICE), IL-1 receptor antagonist (IL-1RA) mRNAs were both lobes. Type (IL-1R1) constitutively expressed pituitary, its level did not change LPS injection. Interestingly, coding glial fibrillary acidic protein (GFAP), an astrocyte marker, selectively suggesting affects pituicyte function. Together, results suggest acts directly on rapidly induce expression. Locally synthesized may activate bearing cells modulate endocrine activities
