摘要: The mouse anterior pituitary contains both types of interleukin (IL)-1 receptors, IL-1 receptor type I (IL-1RI) and II (IL-1RII). These receptors are expressed mainly on somatotroph cells. In the present study, ability to respond in vivo or lipopolysaccharide (LPS) was demonstrated by measuring, with an electrophoretic mobility shift assay, presence active NF kappa B complex cell nuclei from pituitaries mice injected intraperitoneally recombinant rat-IL-1 beta LPS. Using immunohistochemistry antibody directed against p65 subunit, a rapid transient response LPS observed. This predominantly glial fibrillary acidic protein (GFAP)-positive cells F4/80-labelled posterior 15 min after stimulation became faint 2 h. comparison, early strong treatment localized into cells, GFAP positive pituitary. Activation no longer apparent IL-1RI knockout mice, confirming that this is essential for transduction signal pituitary, but remained treatment. addition, we investigated effect target genes measuring mRNA proteins synthesis growth hormone (GH), IL-6 IL-1ra plasma. shown induce failed regulate GH contents release. data suggest able systemic infection via cytokine-mediated responses transduced IL-1.
