摘要: Abstract : In order to identify and characterize microbes using the currently available methods, information on organism's genetic material is required. Recombinant which contain novel would not have such therefore avoid detection. Previous work has been done develop a system with array-based genomic fingerprinting technology, should help new strains, detect presence of material, measure gene expressions. The next phase development test for rapid species identification in addition oligonucleotide fingerprint. organisms this were Bacillus bacteria (11 species), Escherichia coli TOP10 (7 strains), Geobacillus stearothermophilus. Using standard molecular biology we isolated DNA, digested DNA reduce complexity, labelled it fluorescent dyes, hybridized two types microarrays, Human Operon 21K chips containing 23,232 features Bacterial 5,280 features. hybridization data was then analyzed ChromaBlast, an useful analytic tool Excel, normalized columnar data, sorted into user-selectable range-driven bins, developed colour heat maps from output map bin assortment review. When patterns filtered sorted, different genera could be discriminated high confidence certain subsets However, or strain differentiations as evident work. A multipathogen chip designed further investigate differentiation. Due time limitation contract, only one sample each organism included thus far. phase, more replicate microarray set data.
