摘要: The yeast Xanthophyllomyces dendrorhous is used for the microbiological production of antioxidant carotenoid astaxanthin. In this study, we established an optimal protocol protein extraction and performed first proteomic analysis strain ATCC 24230. Protein profiles before during induction carotenogenesis were determined by two-dimensional polyacrylamide gel electrophoresis proteins identified mass spectrometry. Among approximately 600 observed spots, 131 non-redundant identified. Proteomic analyses allowed us to identify 50 differentially expressed that fall into several classes with distinct expression patterns. These demonstrated enzymes related acetyl-CoA synthesis more abundant prior carotenogenesis. Later, redox- stress-related up-regulated For biosynthetic mevalonate kinase phytoene/squalene synthase, higher abundance and/or accumulation carotenoids. addition, classical enzymes, such as catalase, glutathione peroxidase cytosolic superoxide dismutases, not Our results provide overview potentially important carotenogenesis-related proteins, among which are involved in carbohydrate lipid pathways well proteins. these might indicate X. accumulates astaxanthin under aerobic conditions scavenge reactive oxygen species (ROS) generated metabolism.
