Comparison of neurotrophin and repellent sensitivities of early embryonic geniculate and trigeminal axons

摘要: Geniculate (gustatory) and trigeminal (somatosensory) afferents take different routes to the tongue during rat embryonic development. To learn more about mechanisms controlling neurite outgrowth axon guidance, we are studying roles of diffusible factors. We previously profiled in vitro sensitivity axons neurotrophins target-derived factors now report on these properties for geniculate axons. GDNF, BDNF, NT-4, but not NT-3 or NGF, stimulate ages investigated, days 12-14. Sensitivity effective is developmentally regulated from that ganglion. In coculture studies revealed were repelled by branchial arch explants shown be repellent (Rochlin Farbman [1998] J Neurosci 18:6840-6852). addition, some untransfected COS7 cells Sema3A, a ligand neuropilin-1, repelling axons, antineuropilin-1, antineuropilin-2, completely blocks repulsion repel both ganglia. Sema3A mRNA concentrated epithelium at appropriate time mediate repulsion. knockout mice, explore medial regions immature surrounding territories explored heterozygotes, supporting our previous hypothesis Sema3A-based mediates early restriction sensory away midline structures.
