Receptor activation regulates cortical, but not vesicular localization of NDP kinase

摘要: We used immunofluorescence techniques to determine the localization of nucleoside diphosphate (NDP) kinase in NIH-3T3 fibroblasts. found that cytoplasmic NDP can be separated into two populations according subcellular and response extracellular stimuli. Specifically, within minutes stimulation resting fibroblasts with serum, growth factors or bombesin, a portion becomes associated membrane ruffles lamellipodia. Another pool accumulates independently around intracellular vesicles. Transfection cells activated Rac mimics, whereas expression dominant negative inhibits, effects on translocation cell cortex. Neither mutant affects vesicle-associated pool. Association vesicles depends microtubule integrity is disrupted by nocodazole. In cell-free assays binds tightly taxol-stabilized microtubules. Binding this fraction reduced ATP abolished GTP, as well guanine nucleotides are substrates. Thus, pools identified here regulated distinct cellular components: appearance cortical consequence activation, vesicular responsive dynamics nucleotides, particular GTP. These results suggest participates Rac-related events GTP-dependent processes linked vesicle trafficking.
