摘要: Nm23 has been identified as a gene family encoding different isoforms of nucleoside diphosphate kinase (NDPK). This protein is key enzyme in nucleotide metabolism and shown to play important roles various cellular functions. In the present study, we have investigated expression three isotypes mouse dorsal root ganglia. situ hybridization reverse transcriptase-polymerase chain reaction analysis demonstrated high levels nm23-M1, -M2, -M3 mRNA peripheral nervous tissue. Moreover, also displayed specific nuclear localization for nm23-M2 mRNA. Immunohistochemistry with light electron microscopy on isoform-specific antibodies revealed differential subcellular distribution NDPK isoforms. Isoform A was mainly cytosolic, showing only partial association organelles. contrast, isoform B found nucleus, which agreement its proposed role transcription factor. The results indicate preferential C endoplasmic reticulum plasma membranes neuronal cells. Furthermore, appeared combine other by double-labeling evidence might be responsible binding oligomers membranes. Thus, may considered importance maintaining intracellular pools GTP vicinity and, hence, transmembrane signaling. isoforms, not central but system.
