Transcriptomic analyses of genes differentially expressed by high-risk and low-risk human papilloma virus E6 oncoproteins.

摘要: Human papilloma virus is the causative agent for cervical cancer with 99 % of cases containing HPV. The high risk HPV-16, 18 and 31 are major agents. low HPV-6, 11 have been reported to cause penile, laryngeal, bronchogenic oesophageal cancer. Since E6 oncoprotein frequently over expressed in cancers, we did gene expression studies compare between genes high-risk (HPV18) or low-risk (HPV11)stably transfected epithelial cell line EPC-2 mock basic vector pCDNA3.1. Microarray showed a total 697 showing differential samples. Genes involved several key cellular processes such as adhesion, angiogenesis, transcription regulation, cycle regulation division altered Gene Ontology mapping 44 according pathways revealed 13 namely alzhemier’s, Wnt, p53, interleukin, TGF-β, cadherin, integrin, PI3-kinase, catennin, insulin, chemokine G protein signalling pathways. microarray results were confirmed by quantitative real-time PCR some representative like IFI27, CTNNA1, OSMR, CYP1B1, TNFSF13, LAMA2 COL5A3. Analysis differentially HPV proteins might help identification potential biomarkers diagnosis, progression therapy understanding mechanisms activation these well function products will give further insight into their roles
