Keap1-Nrf2 signaling pathway: Mechanisms of regulation and role in protection of cells against toxicity caused by xenobiotics and electrophiles

摘要: The transcription factor Nrf2 governs the expression of a considerable group genes involved in cell protection against oxidants, electrophiles, and genotoxic compounds. activity is sensitive to xenobiotics endogenous electrophiles. negatively regulated by specific suppressor protein Keap1, which also receptor electrophiles adapter for Cul3 ubiquitin ligase. Electrophiles react with critical thiol groups Keap1 leading loss its ability inhibit Nrf2. Keap1-Nrf2 signaling pathway down-regulates NF-κB transcriptional attenuates cytokine-mediated induction proinflammatory genes. Pharmacological activation can be used treatment prevention many diseases. Widely known natural activators include curcumin, quercetin, resveratrol, sulforaphane. most effective are synthetic oleanane triterpenoids.
