Arginine and lysine reduce the high viscosity of serum albumin solutions for pharmaceutical injection

摘要: Therapeutic protein solutions for subcutaneous injection must be very highly concentrated, which increases their viscosity through protein-protein interactions. However, maintaining a solution below 50 cP is important the preparation and of therapeutic solutions. In this study, we examined effect various amino acids on concentrated bovine serum albumin (BSA) human (HSA) at physiological pH. Among tested, l-arginine hydrochloride (ArgHCl) l-lysine (LysHCl) (50-200 mM) successfully reduced both BSA HSA solutions; guanidine (GdnHCl), NaCl, other sodium salts were equally as effective, indicating electrostatic shielding these additives. Fourier transform infrared spectroscopy showed that in its native state even presence ArgHCl, LysHCl, NaCl high concentrations. These results indicate weakened interactions play key role reducing viscosity. ArgHCl are also non-toxic compounds, will used additives to reduce proteins.
